This is at the very end of the story when General Zaroff walks into his bedroom and finds Rainsford waiting for him in there. After a bit of a conversation between the two it becomes obvious there will be conflict, so Zaroff tells Rainsford "On guard,Rainsford...".
In this part, the "hunt" had just begun and Zaroff had tracked Rainsford through all of his attempts to throw him off his path. In the story Rainsford feels the urge to "hurl himself down like a panther", but he has to control himself and sit as still as possible.
In the very beginning of the story, Rainsford ends up falling off of his boat, and is struggling to swim in the water. To quote: "... as the blood-warm waters of the Caribbean Sea closed over his head. ... Desperately he struck out with strong strokes towards the yacht."
