  • Gram Staining Step 1: Heating the Slide
  • Gram Staining Step 2: The Dye
  • Gram Staining Step 3: Rinse
  • Heat fix the slide to ensure that the bacteria cells are fixed to the slide.
  • Gram Staining Step 4: The Iodine
  • Flood the slide with crystal violet dye- The dye ions penetrate the cell wall and membrane.
  • Gram Staining Step 5: Rinse Again
  • Rinse the slide with water.
  • Gram Staining Step 6: Decolorizing with Alcohol
  • Flood the slide with Iodine- the iodine ions penetrate the cell wall and join in with the crystal violet. This reacts and creates a complimentary large molecule that is insoluable in water
  • Rinse the slide with water.
  • Decolorizing the solution with alcohol for gram negative would mean the water would leave the cell and the iodine would be blocked from leaving. In gram positive, the alocohol dissolves the outer membrane, which lets the iodine leave.
