In the Devil in the White City there are two plots running simultaneously. The main characters of the first plot are doctor H.H. Holmes and detective Frank Geyer. The main conflict of this plot is that Holmes' murders go unnoticed while he profits off the victims.
The main characters of the second plot are lead architect of the fair Daniel Burnham, his associate John Root, and landscape architect Frederick Law Olmsted. The main conflict of this plot is that the architects led by Burnham are on a race against time and environmental conditions to finish the World Fair.
The climax of the novel is when Patrick Prendergast an aspiring architect assassinates Chicago mayor Carter Harrison. This event brings the closing of the World Fair.

The setting of the novel takes place in Jackson Park where the fair is to be built and a suburb of Chicago called Englewood. The entire novel is set between the years 1889-1993.
The theme of the novel is duality. The Burnham plot symbolizes sanity while the Holmes plot represents insanity. Likewise the White City is the source of happiness and joy while Holmes' building is the source of evil and crime.
