  • Scientists Working in a Lab
  • Hey Baylee, what do you want to observe or test today?
  • Ok, then.
  • I'm not sure yet Brandon, why don't we look outside?
  • Brainstorm testing Ideas
  • Alright, lets test it!
  • Why don't we see what's occurring in the soil over there?
  • You'll Never find me!
  • Collect Soil Samples
  • NOOOOO!
  • Testing for Dehydrogenase Activity
  • What does that mean?
  • Woah! The soil tested positive for Dehydrogenase activity.
  • Dehydrogenase activity in soil oxidizes a substrate by moving hydrogen to an acceptor (receiver) that is either NAD or NADP.
  • However some factors can affect dehydrogenase activity.
  • 1. Temp. of Environment Affecting Dehydrogenase Activity
  • A factor that can affect Dehydrogenase activity is environmental factors such as temperature. Dehydrogenase activity can complete reactions efficiently when temperature is 20-30 degrees celsius but also stable enough for the microorganisms as well.
  • Wow! Whats another affect?
  • 2. pH affecting Dehydrogenase Activity
  • Hmm, I'm starting to get it more and more. What is another factor?
  • Another factor that can affect dehydrogenase activity is pH level. If the pH level is not between 5.5-5.7, then dehydrogenase cannot perform reactions properly.
  • I didn't know that. Is dehydrogenase activity play a part in any biochemical cycle?
  •
  • 3. Trace Elements affecting Dehydrogenase Activity
  • The last but not least factor that affect dehydrogenase activity is trace elements in soil. trace elements in soil such as alkaline metals can greatly affect the activity.
  • When testing for soil, triphenyltetrazolium chloride can be used as an indicator to find the presence of dehydrogenase activity. If the soil substance and triphenyltetrazolium chloride is red, the test is positive for dehydrogenase activity.
  • Involved in Carbon and Phosphorus Cycle
  • Yes. Dehydrogenase activity is involved in photosynthesis thus being in the carbon and phosphate cycle.
  • CONTINUATION: A factor that can affect Dehydrogenase activity is environmental factors such as temperature. Dehydrogenase activity can complete reactions efficiently when temperature is 20-30 degrees celsius but also stable enough for the microorganisms as well. However dehydrogenase activity decreases during colder seasons due to a decrease in temperature.
  • Since, my temperature is less than 20-30 degrees Celsius, I can't complete reactions as much.
  •
  • OH NO! My pH level is less than 5.5-5.7 so I can't perform activities properly.
  • OH NO! My pH level is over 5.5-5.7 so I can't perform activities properly.
  •
  • Oh no! These alkali metals are going to denature me!
  • We Alkaline metals will denaturize you dehydrogenase by by interacting with the enzyme-substrate complex,
  • CONTINUATION: In photosynthesis, a Gunam absorbs sunlight and H2O Carbon Dioxide is also absorbed. Dehydrogenase activity transfers Hydrogen from NADPH so that it is used in carbon dioxide to make sugar for the plant as C6H12O6.
  • Amazing!
  • ADP+P+NADP
  • ATP+NADPH
