Charlie Gordon is an unintelligent man who wishes to be the contrary and so, takes part in an experiment for that. He first takes the Rorschach test but is unable to see the "pictures" in the inkblot.
Charlie does a maze along with a mouse named Algernon doing the same maze which results in Charlie losing to Algernon.
After the operation and several more mazes, Charlie finally beats Algernon in a maze.
After becoming intelligent and learning and experiencing many things, Mr. Donnegan, the boss of Donnegan's Plastic Box Company, shows Charlie the petition to have him fired from his job.
Charlie writes a report titled "The Algernon-Gordon Effect" which is about how the operation that made Charlie more intelligent rapidly also reduces it in the same amount of time.

After losing his intelligence and Algernon's demise, Charlie moves away from New York to a place where he will not be recognized as once being intelligent and takes along some books.
