When Iago commands Emilia to go steal the handkerchief, Emilia right away goes to steal it. As easy as it was Desdemona doesn't realize it has fallen to the ground and takes off with Othello giving Emilia an easy shot in getting the handkerchief.

Here is where Emilia gives the handkerchief to Iago, trying to get the reason why he commands her to steal it but Iago refuses to tell her and tells her to leave .

Later on Iago lies to Othello stating that he has seen the handkerchief with Cassio, This handkerchief is important because it was a gift from Othello's mother once she died. Othello gave this gift to Desdemona symbolizing her loyalty.
