It would add another slave state to the union and it would let slavery spread to the west. It would be harder to abolish and would disrupt the balance in Congress.

Northerners got more land!
They got Missouri out of it and they thought that they would get California, New Mexico, and Texas sooner or later, meaning more slave states.
We're going to get Texas and California Eventually!
They thought that states should decide whether they want slavery or not. It is not up to the federal government. Calhoun believed that the states had the right to decide.
Federal government shouldn't decide!
The Northerners ended up gaining more land from the compromise, making the southerners mad. They will get a majority and outnumbered the amount of slave states, hence, abolishing slavery. Also, they won't be able to farm cotton cheaply anymore.
